Studies on Toxigenicity of Verotoxin-producing Escherichia coli O157

Abstract
Verotoxin produced in culture was quantified by RPLA method for 34 Escherichia
Coli O157 strains isolated from human and environmental materials in Miyazaki
Prefecture in 2000-2003.
As a result, VT1 was produced in bacterial culture with
CAYE medium(CAYE) more in quantity than Trypto-soya broth(TSB), and was caused
significant increase by Polymyxin B treatment. On the other hand, VT2 was produced
in almost same amount in each culture with CAYE and TSB, and was caused no increase
by Polymyxin B treatment.
Among all tested strains, little difference of quantity
was showed in VT1 production, but significant difference was showed in VT2 production.
From
the quantitative analysis of verotoxin of EHEC O157 strains isolated from adults,
symptoms seems to be caused by the strains which showed high toxigenicity of
VT2.

はじめに
平成15年4月、宮崎市において、腸管出血性大腸菌(EHEC)O157に感染した小児2名が死亡する事例が発生した。EHECO157によって引き起こされる症状は、HUSや脳炎を併発し重症化するものから無症状のものまで、その程度は様々である。このような症状の違いは、感染菌量、ベロ毒素(VT)の種類や産生能、また宿主の年齢、基礎疾患の有無等が関与していると考えられる。そこで、今回、症状の差に関与する因子の一つとして毒素産生能について検討するため、宮崎県で分離されたEHECO157菌株の試験管内培養での毒素産生能を比較し、症状との関連性をみることを試みた。
